This sequencing was designed to identify the downstream genes that were regulated by CRY2 in darkness and blue light or regulated by FL1/FL3 in darkness. For the comparison between WT and cry2 mutant, 4-day-old seedlings grown either in darkness or transferred from darkness to blue light for 30 minutes were harvested and rapidly frozen in liquid nitrogen. Subsequently, the frozen samples were used for mRNA-seq analysis. For the comparison between WT and flΔ4 mutant, 4-day-old seedlings grown in darkness were harvested and rapidly frozen in liquid nitrogen. Subsequently, the frozen samples were used for mRNA-seq analysis.
