Through a genetic screen aimed to identify host factors for LTR-retrotransposons, we identified the pivotal role of two interacting translesion DNA polymerases, Rev1 and PolH. These associate with RNA Pol II, and facilitate Pol II occupancy at LTR-retrotransposons, thereby boosting LTR-retrotransposon transcription in both the germline and somatic tissue.
