A plasmid library containing all possible single codon substitution and deletion mutations to the AAV2 rep gene was created. All library variants include unique 20 base pair barcodes. The effect of each rep variant on AAV2, AAV5, and AAV9 capsid production was measured via multiplexed transfection and subsequent barcode sequencing.
