Tumor associated macrophages (TAMs) play a critical role in biology of various cancers. TAMs are known to polarize to either M1, which represents anti-tumor activity, or M2, which leads to tumor promotion. Numerous studies have shown that M2 TAMs promote tumor occurrence and development by secreting vascular endothelial growth factor (VEGF), which is then involved in angiogenesis. Recently, the development of TAMs in vitro has been used to generate preclinical models for cancer research. However, the similarity between in vitro and in vivo TAMs are still poorly understood. Here, we provided a better model for the generated of TAMs. We discovered in vitro-generated TAMs by culturing RAW264,7 macrophage cell lines in tumor conditioned media (TCM) derived from 4T1 murine mammary carcinomas.
