The U2OS cells were obtained from the American Type Culture Collection (ATCC) and additionally genetically authenticated by DKFZ (Heidelberg) core facility. The cells were treated either with different concentrations of 5-Aza-2′-deoxycytidine (decitabine, DAC) in culture or control DMSO containing media (corresponding to its concentration in the 10 µM DAC treated cells). Cells were harvested after 96 hours incubation and used for RNA and DNA extraction.
