Methylobacterium strains (Bacteria, Proteobacteria, Alphaproteobacteria, Rhizobiales, Methylobacteriaceae) isolated from leaf samples from two forests in the province of Quebec, Canada: Mont Saint Hilaire (MSH) and Station Biologique des Laurentides (SBL). Strains were isolated on minimum salt media with methanol 0.1% as sole carbon source. Each tree was sampled either once in August 2017 in MSH (19 trees, pilot survey), or 3-4 times from June to October 2018 in both MSH and SBL (52 trees, seasonal survey). (i) 80 strains and identified by Sanger-sequencing of the 16S V4 region. (ii) 20 strains isolated from the pilot survey used to develop a Methylobacterium-specific marker by Sanger-sequencing of three and two hypervariable regions of genes sucA and rpoB, respectively. (iii) 167 strains isolated from the seasonal survey and identified by Sanger sequencing of rpoB.
