Expression data from control and CTIP2-/- (BCL11B) hESC-derived medium spiny neurons. Homo sapiens

Overall design: CTIP2 knock-out hESC line was generated using the CRISPR/Cas9-assisted genome editing technology with guide RNAs targeting exon 2 of the CTIP2 gene. To investigate whether CTIP2 deficiency affects the generation of MSNs from hESCs, we performed striatal neural differentiation on the CTIP2-/- line along with the isogenic control HUES9 iCas9 cells using an established protocol (Arber et al., Development 2015). Total mRNA from control and CTIP2-/- neurons was collected and prepared for RNA-seq analysis at day 20 and 40 in vitro neural differentiation.
