Slfn2 Regulates Type I Interferon Responses by Modulating the NF?B Pathway

We demonstrate that Slfn2 interacts with protein phosphatase 6 regulatory subunit 1 (PPP6R1), leading to reduced type I IFN-induced activation of nuclear factor kappa B (NF?B) signaling, resulting in reduced expression of ISGs. Overall design: Slfn2 WT and Slfn2 KO mouse embryonic fibroblasts (MEFs) were plated in triplicate/duplicate and were left untreated or were treated for 6 hours with 5x103 IU/ml of mouse IFNß, followed by total RNA isolation. Directional libraries were prepared using Illumina TruSeq Stranded Total RNA HT Sample Prep Kit.
