General PLS3 KO mice model was generated using the CRISPR/Cas9 system at Cyagen Biosciences (Suzhou) Inc. We performed one times in vivo axial loading on 26 mice (WT male: 11; Hemizygous: 15), 5 WT male mice and 5 PLS3 KO male mice bone mRMA were selected through blind selection. Under anesthesia, the right tibias were loaded to a peak force of 8N for 40 cycles/day; contralateral left tibias served as non-loaded control. Electropulse 3000 system (Instron, USA) was used to apply axial compression. Then total 20 samples from 10 mice were used for RNA-seq.
