Indian River Lagoon Water Prokaryotic Communities Survey 2016-2018

This study was conducted to act as a baseline assessment of the water microbiome of the Indian River Lagoon, FL, USA an Estuary of National Significance. Water was collected during the same sampling trips as the sediment samples in the previously published study on sediment (PRJNA594146 ). Extracted DNA samples were sent to Research and Testing (Lubbock, TX, USA) for MiSeq 16S sequencing to amplify the bacterial/archaeal 16S V4 region using the modified primers used by the Earth Microbiome Project of 515F (GTGYCAGCMGCCGCGGTAA) and 806R (GGACTACNVGGGTWTCTAAT). It is a two-year survey of 11 sites taken during four sampling periods about six months apart at locations present to continuous water quality monitors. The primary goal of this sampling was to complement the sediment samples and assess the impact of freshwater discharges on the water microbiome.
