The kidney is a heterogeneous organ with many types of cells, and therefore, it is difficult to clarify the cell-specific changes in molecular events. To overcome this problem and examine the gene expressional profile localized in the renal proximal tubules accompanied by progression of chronic real failure, rats were subtotal nephrectomized (Nx) and the renal proximal tubules were isolated with microdissection technique in serial time points after Nx. We conducted microarray analyses with the isolated proximal tubules and clearly demonstrated time-dependent changes in gene expression profiles. The total numbers of genes that significantly changed more than 2-fold compared to the control at 1, 2, 4, and 8 weeks after surgery were 239, 161, 278, and 389, respectively. Using the total number of genes, we classified them according to their Gene ontology using the MetaCoreTM software to identify the biological function involved in the progression of CRF in the proximal tubules. The results revealed that the cell cycle or cytoskeleton-related genes were significantly altered at 1 and 2 weeks after Nx. Additionally, inflammation-related genes were frequently observed at 8 weeks after Nx.
