PI3KCD Gene: A Key Player in Diabetic Intervertebral Disc Health

Objective: To study the promoting factors of intervertebral disc degeneration in patients with diabetes;Methods:Collect IVDD data of ordinary patients and diabetes patients for statistical analysis.After culturing nucleus pulposus cells with different high glucose concentrations for 3 days,WB and RT-qPCR was performed to detect the degeneration markers of nucleus pulposus. Transcriptome sequencing was conducted on both the control and experimental groups and the target PI3KCD was identified. SiRNA and plasmids were used to knockdown and overexpress gene PI3KCD. WB and RT-qPCR were used to redetect the degeneration markers. The PI3KCD agonist 740Y-P was added to the nucleus pulposus cell culture plates, WB and RT-qPCR were used again to detect the degeneration markers. The agonist 740Y-P was injected into the intervertebral discs of rats. After one week, the intervertebral discs were collected to prepare tissue sections, which were immunofluorescence stained and imaged to detect the degeneration markers.Results:Transcriptome sequencing under high sugar environment,PI3KCD significant decrease,PI3K-AKT signaling pathway has undergone significant changes.Knockdown and overexpression of PI3KCD lead to changes in degeneration indicators of NP cells.Conclusions: This study confirmed that high glucose decreased the expression of the PI3KCD gene, which in turn sped up the breakdown of intervertebral disc tissues through the PI3K/AKT pathway.
