Images of lysotracker fluorescence (with Hoechst counterstaining) analysed by lysosomal area measurement in ImageJ and Lysotracker fluorescence analysis by fluorescence plate reader in CHO cells (control CHO H1 and also Niemann-Pick C disease CHO M12) grown in different concentrations of HEPES buffer (over short and long term) and different zwitterionic buffers. Further data in iPSC-neurons grown with and without HEPES for 1 week with images of lysotracker fluorescence (and phase contrast) and analysis of lysosomal Ca2+ levels by Fura 2 in neurons (grown in media either with and without HEPES) treated with ionomycin and then GPN to specifically release lysosomal Ca2+.
