We performed single cell RNA-sequencing (scRNA-Seq) of testes from bovine fetuses derived from either CRISPR/Cas9 NANOS3 knockout (KO; NANOS3 -/-) or wildtype control (NANOS3 +/+) embryos. The scRNA-Seq analysis showed a complete loss of primordial germ cells (PGCs) and gonocytes in NANOS3 KO fetal testes, while maintaining the development of somatic support cells. Overall design: Bovine fetal testes derived from NANOS3 knockout and wildtype embryos were collected and prepared for scRNA-Seq analysis. Libraries were prepared with the whole transcriptome 100K kit from Parse Biosciences, which involves barcoding and then pooling cells from all samples prior to the generation of eight sublibraries. Because of the kit's design, sequencing reads of all samples are distributed across all eight sublibraries. For further details see the attached `README.tsv` file.
