Sex reversal phenomena are observed in natural populations of teleost, with the Monopterus albus serving as an ideal model for studying sex determination. In this study, we have employed a single cell RNAsequencing approach using 10xGenomics scRNAseg to characterize the complex cellular compositions of the ovary ovotestis, and testis, and traced their cellular and molecular transitions during sex reversal. We have not only provided a detailed depiction of the differentiation of germ cell lineages but also offer thefirst characterization of different functional subpopulations of somatic cells in the Monopterus albus gonad, revealing their roles in sex hormone synthesis and their fates during sex reversal.
