SbBES1 was firstly expressed in sorghum leaves by agroinfiltration, to ensure proper expression and localization of the recombinant protein. The sorghum BES1 protein was localized in the nuclei and the cytoplasm, in accordance with previous studies performed in the plant model system Arabidopsis (Ryu et al., 2010). After validation of the subcellular localization of sorghum SbBES1, DAP-seq experiments were performed to identify the putative SbBES1 direct targets by mapping the sequencing data in the Sorghum bicolor v3.1.1 (Phytozome genome ID:454) reference genome. A total of 2255 peaks were detected with a significant enrichment compared to the Halo-input libraries, and the nearest genes were considered as SbBES1-bound. Binding peaks were selected in the base of bell shape peaks, the enrichment against the Halo-input signal and the presence of a motif, analyzed by motif k-mer identification in MEME database. After performing motif enrichment on the TOP600 best-scoring SbBES1 bound genes, we observed the enrichment of a G-box (CACGTG) element.
