Select chromatin complexes are essential drivers of pathogenic enhancer activity and gene expression in Ewing sarcoma [ChIP-Seq]

Using ChIP-seq we assessed the changes occuring at EWS::FLI1 enhancer regions following perturbation of EWS::FLI1, MLL3/4 and p300/CBP. Overall design: Using previously described EWS::FLI1 degrader cell lines (A673-dHALO and EWS::502 dTAG) we assessed the changes in the epigenetic landscape at EWS::FLI1 enhancers compared to MLL3/4 genetic deletion and p300/CBP perturbation using small molecule inhibitor A485 and degrader dCBP1.
