We performed RNA sequencing to identify the role of fibroblast in the effects of tryptphan metabolites on LPS-stimulated RAW264.7 cells. Overall design: Intestinal fibroblasts were cultured and passaged into 24-well plates. Separately, RAW264.7 cells were passaged and seeded into 0.3 µm pore-size transwell inserts, which were then placed into the 24-well plates containing the fibroblasts. The co-cultures were then exposed to lipopolysaccharide (LPS, 100 µg/mL), indole lactic acid (ILA) or indole acetic acid (IAA) (each at 6 µM), and/or CH223191 (1 µM), depending on the experimental group.
