We performed in vivo CRISPR screen of Toxoplasma targeting ROP/GRA genes to identify in vivo fitness genes in WT and Ifngr1-deficient mice. This sublibrary targeting 376 genes which consisted 3757 gRNAs targeting 376 putative or known ROP/GRA and RON genes together with dispensable and essential control genes. We performed three independent in vivo CRISPR screens to identify novel virulence factors of Toxoplasma.
