Naïve, central memory and effector memory CD4 T cells from healthy young and older adult individuals were purified by cell sorting for CD62L, CD28 and CD45RA. For ATAC-seq, sorted cells were resuspended in the transposase reaction mix (25μL 2× TD buffer, 2.5μL transposase (Illumina) and 22.5μL nuclease-free water) and incubated at 37C for 30 min. DNA was purified using Qiagen MiniElute kit and amplified using Nextera PCR primers. Libraries were quantified using a KAPA Library Quantification Kit and sequenced on an Illumina HiSeq 2500. Reads mapped to autosomes are reported and have been filtered for duplicates
