ZymoBIOMICS Microbial Community DNA Standard

500 ng of ZymoBIOMICS Microbial Community DNA Standard (Zymo Research, Cat. #D6306) was fragmented using a Covaris E220 and Illumina library were prepared using NEBNext Ultra DNA II Library Preparation Kit with recommended protocols (New England Biolabs, Cat. #E7645L). Illumina libraries were eluted in DNA Elution Buffer (Zymo Research, Cat. #D3004-4-10). The concentration of the amplicon pools were obtained using the Qubit dsDNA HS Assay (ThermoFisher Scientific, Cat. #Q32854). The average size of the library was determined by the Agilent High Sensitivity DNA Kit (Agilent, Cat. #5067-4626). An accurate library quantification was determined using the Library Quantification Kit - Illumina/Universal Kit (KAPA Biosystems, KK4824). The library was sequenced on the Illumina MiSeq generating paired-end 301 bp reads using approximately 43% of the MiSeq Reagent Kit v3 (600-cycle) (Illumina, Cat. #MS-102-3003).
