In this experiment the relationship between SOX2 levels and the binding of B-catenin, TCF/LEF factors, and SOX2 to chromatin was assayed using ChIP-seq during embryonic stem cell differentiation and in naive pluripotency. A doxycycline inducible cell line (SOX2 TetON) was employed to experimentally control SOX2 expression. Triplicate chromatin immunoprecipitations from independent biological replicates were performed for each factor analysed in four experimental conditions. Sox2 TetON ES cells differentiated in the presence or absence of 1ug/ml doxycycline were compared to differentiated WT epiblast progenitor cells and naive pluripotent embryonic stem cells.
