Genome-wide reciprocal hemizygotes

The goal of this study was to map the genetic underpinnings of trait variation between two species of Saccharomyces yeast. We mutagenized a hybrid of S. cerevisiae and S. paradoxus with a transposon, generating a genome''s worth of reciprocal hemizygotes. This pool was grown at high temperature and a control temperature. The resulting samples were subjected to deep sequencing to assess mutant fitness as a way to map the genes underlying thermotolerance in S.cerevisiae.
