Transcriptional profile of full thickness skin or epidermis only obtained from K14-CreER+/+;GPX4flox mouse model vs K14-CreER-/-;GPX4flox controls is analyzed and compared to transcriptional profile of epidermis plus papillary dermis obtained from patient-matched lesional vs perilesional psoriatic skin. GPX4 was selectively deleted in a subset of K14+ keratinocytes of 8-10 week-old B6.K14-CreER+/+;GPX4flox male and female mice using tamoxifen injections. On day 15-18 after the initiation of tamoxifen injections, either full thickness skin (epidermis and dermis, 3 mice per group) or epidermis only (3 mice per group) were harvested from K14-CreER+/+;GPX4flox mice and K14-CreER-/-;GPX4flox control mice, and used for RNA-sequencing. In addition, clinical specimens composed of epidermis and superficial dermis were obtained from three patients suffering from psoriasis. Specifically, 3 lesional (from 2 males, 1 female) and 3 patient-matched perilesional (located 8 mm from the active lesion) specimens were obtained from the back, thigh or leg, and used for RNA-sequencing.
