Sucrose-induced stolon initiation in vitro potato: integration hormone and RNA-seq analysis

Potato is considered the highest productive crops, and the stolon determine tuber and mini-tubers production. The purpose of the study was to focus on illustrate the action mechanism of potato stolon initial via sucrose induced. An effective inducing model to study sucrose-induced stolon in potato; transcriptomics and endogenous plant hormones were incorporated in present study. Treatment of potato seedings with 8% sucrose in the dark induced the stolon initiation. No stolon was produced for the stem node in the 3% sucrose treatment. Results showed that low-sucrose caused the auxin and CK-related genes lower expressed in stem node and influenced the reductions on content of auxin and iPT, zTR; enhanced the GA-related genes higher expressed in same part, and increase on content of GA3. Interestingly, higher concentration sucrose impressed auxin and ck-related genes highly expressed in the stem node and stolon initial and increased the content of IAA and CK, but GA-related genes lower expressed in the stem, higher in the stolon initial, and the result was consistent with GA3 content in same part. Taken together, our results suggest that sucrose induced stolon is partly promoted by regulating the seven genes auxin regulated protein, GID1, zeamatin, auxin efflux carrier component (PIN6), auxin efflux carrier family protein (PIN), auxin response factor (5,9), causing increased on IAA, CK, GA3.
