To investigate the activation and polarisation of BMDMs during the course of pancreatitis, BMDMs were co-incubated with CCK-stimulated acinar cells (+/- the serine protease inhibitor nafamostat) for 6h in an in vitro experiment. The LPS treatment served as a positive control. Overall design: We used BMDMs isolated from C57Bl/6 mice which were maintained in the presence of 20ng MCSF over 7d until they were confluent. Co-incubation was performed with freshly prepared acinar cells from C57Bl/6 mice over 6h +/- Nafamostat. BMDMs were carefully washed before RNA Isolation. Transcriptome analyses were performed using individual RNA samples.
