A general practical approach toward P(O)Me2–alkenes from ketones has been elaborated. The obtained compounds were modified to provide various P(O)Me2 building blocks for medicinal chemistry. Incorporation of the P(O)Me2 substituent into the antidiabetic drug Gliclazide provided an analogue with improved solubility and reduced lipophilicity.
