RNAseq analysis of NF1 loss of function in immortalized, nontumorigenic human breast epithelial cell line MCF10A. CRISPR guides targeting exon 21 within the GRD domain were constructed and used either as single or dual-guide RNAs with Cas9 nuclease. We established three cell lines (i.e., g1, g2, g1/2) and NF1 loss was validated by PCR and Western blot analysis (Figure S6B). 15 samples derived from the human MCF10 cell line. The samples are divided into 5 genotypes, each with 3 samples. The genotypes are parental MCF10, empty vector control, and three NF1 mutants: guide RNA 1, guide RNA 2, and guide RNA 1 and 2.
