Cleavage Under Targets and Release Using Nuclease (CUT&RUN) was performed using CUTANA protocol v1.6 [www.epicypher.com]. Human pulmonary arterial smooth muscle cells (hPASMCs) were treated with vehicle, EMAP II or EMAP II+ SPHK2 inhibor and nuclei from two biological replicates were extracted. For each target 500K nuclei were immobilized onto Concanavalin-A beads and incubated overnight with of antibody IgG (negative control) or Ac-H3K9 (all validated to SNAP-ChIP nucleosome standards).
