Human umbilical cord-derived mesenchymal stromal cells (UC-MSCs) were rejuvenated by Sendai virus (SeV)-mediated, non-integrative, temperature-sensitive, transient expression of hTERT, BMI1, and SV40T. The rejuvenated MSCs (rej-MSCs) exhibited enhanced proliferative capacity, telomere elongation, and no detectable chromosomal abnormalities. Stranded mRNA-seq was performed on RNA samples derived from SeV-transduced UC-MSCs to (i) confirm safe and complete SeV clearance, (ii) delineate gene-expression trajectories associated with rejuvenation, (iii) identify reliable molecular hallmarks of partial reprogramming, and (iv) evaluate therapeutic relevance and clonal diversity for future MSC banking and precision applications.
