To determine the transcriptomic effects of cold exposure on skeletal muscle gene expression in rats with and without UCP1, we assessed gastrocnemius muscle in chow-fed control and whole body UCP1 knockout rats using RNA-seq. Gastrocnemius muscles were dissected from 7 male control and 9 male UCP1 knockout rats. Total RNA was prepared using Trizol, RNA concentration was determined usign a nanodrop UV spectrophotometer, and 500ng RNA per sample was given for sequencing library construction.
