This experiment was designed to study the effect of the histone H3.3-G34R mutation on the transcriptome of tumor cells and immune-infiltrating cells, in a high grade glioma mouse model. High grade gliomas (HGG) were generated using the Sleeping beauty system and involved these combinations: (i) pSB, pNRAS-G12V, pshATRX and pshp53 (H3.3-WT group), (ii) pSB, pNRAS-G12V, pshATRX, pshp53 and pH3.3-G34R (H3.3-G34R group). There are four groups with 3 or 4 samples each, allowing two independent pairwise comparisons: tumor with overexpression of mutated histone H3.3 G34R (n=3) vs H3.3-WT tumors (n=4), and imune cells isolated from H3.3-G34R tumors (n=3) vs immune cells isolated from H3.3-WT tumors (n=4)
