资源简介:
Here we characterize human fetal cardiac lymphatic vessel development and reveal a new role for cells that reside in the epicardium as a source of progenitors involved in cardiac lymphatic differentiation. The single nuclei (sn) Multiome data of the human heart reveal the plasticity and the heterogeneity of the cardiac lymphatic endothelium. We have captured nuclei from a novel population of lymphatic progenitors in the epicardium and performed an integrative snRNAseq and snATACseq analysis to define the gene expression and gene regulatory landscape of these cardiac lymphatic cells in human fetal hearts. Overall design: Micro-dissection of the epicardial layer of the human fetal hearts surface was performed at post-conception week 10 and 11 before lysis of the nuclei and processing for 10x Genomics "Chromium Single Cell Multiome ATAC + Gene Expression". Expression profiling by high throughput sequencing and Genome binding/occupancy profiling by high throughput sequencing from 2 different time points 10 PCW (n=2) and 11 PCW (n=3). ATAC-seq data not provided due to patient privacy concerns
