Iron deficiencies are the most common non-enteric syndromes observed in patients with inflammatory bowel diseases (IBD), yet little is known how iron deficiencies impact immune tolerance in the intestine. The goal of this study is to illuminate role of iron in colonic Treg homeostasis regulation. To investigate which subsets of Treg cells were strictly dependent on TfR1-mediated iron uptake, we conducted single-cell RNA seq (scRNA seq) analysis of total Treg cells (CD25+CD4+) isolated from heterozygous Tfrc fl/fl Foxp3 YFP-IRES-Cre mice ,TfR1-deficient cells (YFP+) and WT (YFP-) cells were separated in silico based on detection of the YFP-IRES-CRE transcript.
