Full-length transcriptome sequencing of the right ventricular myocardium in the control group and high-frequency pacing heart failure canines

Six beagles were randomly divided into the control group (n=3) and the heart failure group (n=3). The control group underwent a sham operation, and the right ventricular pacemaker implantation was used to construct a dog model of heart failure. Heart failure dogs were treated with 3.2 million IU of penicillin + 100 ml 0.9% NaCl intravenously for 2 weeks. After that, the pacemaker was turned on at 180 beats / min for 3 days; 220 beats / min for 3 days; then 250 beats/min for 3 weeks. After the heart failure was confirmed by echocardiography, the right ventricular myocardium was isolated and RNA was extracted. The PCR products were then subjected to Oxford Nanopore Technologies (ONT) adaptor ligation. The final cDNA libraries were added to FLO-MIN109 flowcells and run on PromethION platform.
