Promoter CpG content regulates DNMT-dependent silencing dynamics

Effect of how CpG density on the promoter affects the gene silencing and DNA methylation rate of a promoter We constructed a library of promoter based on the synthetic pEF1s promoter, with varying CpG density. Then we recruited an ectopically expressed DNMT3b to the locus. The cells are sorted into high, med and low expression at different day after DNMT3b recruitment, and subjected to targeted, methylation specific, enzymatic based sequencing analysis (NEB EM-Seq).
