Genome wide RNA expression profiles of endometrial stromal cells stimulated by cAMP, cAMP+MPA, MPA and E2+MPA.

To know the differences of decidualization induced by different stimuli, we compared the mRNA expression profiles of decidualized ESCs stimulated by cAMP or cAMP+MPA for 4 days, or by MPA or E2+MPA for 14 days. ESCs were treated with cAMP or cAMP+MPA for 4 days or, MPA or E2+MPA for 14 days.Because the cultured period was different between the 4-days protocol and the 14-days protocol, we prepared two control ESCs that were cultured without stimulations for 4 days or 14 days, respectively. The mRNA-seq libraries were generated. After the quality of the library was validated using the Agilent Bioanalyzer 2100 (Agilent Technologies, Santa Clara, CA), the samples were subjected to sequencing with a next-generation sequencer (Genome Analyzer GAIIx, Illumina, San Diego, CA). Gene expression values were calculated as “transcripts per million” (TPM).
