Project Objective: The project aims to establish a supportive scaffold for restoring functional tissue after skin injury by developing an engineered cell-scaffold biointerface using a poly-(L-lactide) (PLLA) scaffold enriched with graphene oxide (GO) and gold nanoparticles (AuNPs).Scope of Data: The project includes data on microstructural (SEM), elemental (XPS), thermal (TGA+DSC), and mechanical properties of the scaffolds, along with in vitro assessments of biocompatibility (XTT, LDH assay), adhesion (confocal microscopy, SEM, real-time PCR), and regenerative capabilities (protein membrane, histology) using human keratinocytes (HEKa), fibroblasts (HFFF2), and reconstructed epidermis (EpiDerm™).Tasks Completed:Fabrication of PLLA scaffolds enriched with GO and AuNPs.Comprehensive characterization of the scaffolds’ properties.In vitro evaluations of biocompatibility and regenerative capabilities.Analysis of cellular responses, including morphology, adhesion, and growth factor secretion.The findings suggest that the GO-Au composite improved scaffold properties, enhanced cell behaviors, and has potential in skin tissue engineering to regulate cellular fate and prevent hypertrophic tissue formation.
