RNA-seq in Colias erate

A transcriptome data obtained by RNA sequence (RNA-seq) from a mixture of total-RNA extracted from one male and one female pupae in eastern pale clouded yellow butterfly, Colias erate. We acquired 100-base-paired reads using Hiseq 2500 (Illumina) and conducted de novo assemble to produce contig data by Trinity. The sequence information was further used to design single tagged site (STS) primer pairs and then selected the bacterial artificial chromosome (BAC) clones containing Bombyx mori orthologs. We extracted the BAC-DNA to make probes for BAC-FISH mapping in C. erate.
