Brassica carinata , n:17, genome size :1200 Mb, represent the BBCC genome of Brassicaceae family. We report a chromosome-scale genome assembly of B. carinata var HC20, using a combination of Oxford Nanopore and Illumina sequencing, and BioNano optical maps, resulting in a genome assembly of 1.11 Gb with a scaffold N50 of 39.8 Mb. The chromosomes were developed using a highly saturated genetic map of B. carinata, which were developed using GBS and chip based SNP markers. Two pseudochromosomes contained only one super-scaffold, nine pseudochromosomes contained two super-scaffolds, and the remainder could be assembled with three or more super-scaffolds. The size of the final B. carinata genome that could be assigned to the pseudochromosomes was 1040 Mb. The variety HC20 is highly tolerant to most pathogens of the Brassicaceae family and could a good donor for disease resistance genes to other susceptible Brassica species. This study is a valuable addition to the genomic resources of the Brassica genus and provides a comprehensive strategy to utilize the genomic resources for transferring disease resistance from B. carinata to other high-yielding albeit susceptible Brassica species.
