To explore the existence and distribution of microbiome in tumor tissues and adjacent non-tumor tissues of children with hepatoblastoma (HB), we collected hepatoblastoma tumor tissue and adjacent non-tumor tissue of the patients (n=16) during hepatobiliary surgery. Microbiome was presented in both tumor and adjacent non-tumor confirmed by using qRT-PCR, DDPCR and FISH. The diversity and abundance of microbiome were significantly different between the HB tumor and adjacent non-tumor. At the level of phylum, the dominant microbiomes were Proteobacteria, Bacteroides, and Firmicutes. at the genus level, the abundance of Ruminococcus and in tumor was higher than that in adjacent non-tumor. Simultaneously, the abundance of Bacteroides, Parabacteroides, Lachnospiracea-NK4A136 and Alistipes in tumor was lower than that in adjacent non-tumor. In addition, Romboutsia had a strong correlation with alpha-fetoprotein (AFP), an important indicator of HB. Therefore, it was revealed that microbiome was correlated the occurrence and development of HB. Ruminococcus and Romboutsia were screened as potential bacterial markers of HB tumor. Sphingomonas was rich in primary HB tumor, and Oscillibacter and Pandoraea were rich in metastatic HB. However, if these bacteria were associated with HB should be proved further.
