RNAseq analysis of mouse CD4+ regulatory Tregs in 21-day-old male WT and ∆Foxp3 mice isolated from spleen and lungs. Mouse CD4+ regulatory Tregs in 21-day-old male WT and ∆Foxp3 mice were isolated from spleen and lungs by flow cytometry, as DAPI–TCRβ+CD4+GFP+CD25+. 1,000 cells were double-sorted directly into lysis buffer for RNA-seq (Smart-seq2) library preparation following the standard ImmGen low-input protocol (www.immgen.org).
