Transcriptome sequencing of human liver tissue prior to transplantation

RNA sequencing was performed on liver tissue collected prior to transplantation to profile gene expression in patients who later developed acute rejection and those who did not. Total RNA quality was assessed using an Agilent 2100 Bioanalyzer to ensure high integrity. Libraries were prepared using the QuantSeq FFPE 3'-mRNA-Seq Library Prep Kit FWD (Lexogen) and sequenced on an Illumina NextSeq 2000 platform with paired-end reads. Sequencing reads were quality-checked, adapter-trimmed with TrimGalore, aligned to the human reference genome using STAR, and quantified using Salmon. Gene expression levels were normalized and analyzed for differential expression with DESeq2. This dataset provides insights into pretransplant hepatic gene expression patterns associated with acute rejection risk.
