In our research, we observed that cells undergoing apoptosis emit a red fluorescence when excited by 1060 nm two-photon excitation. This fluorescence has an emission spectrum centered at 600 nm. The enhancement of this fluorescence serves as a novel marker for cell apoptosis, characterized by its non-labeling and non-invasive properties.We discovered that this fluorescence originates from the lysosomes of the cells. Through in vitro experiments, we found that DNA fragments can emit the same red fluorescence under similar two-photon excitation conditions. Therefore, we hypothesize that the intracellular fluorescence is derived from DNA fragments.To test this hypothesis, we extracted DNA fragments from the lysosomes of apoptotic human 231 cells and sequenced these fragments. The data obtained from this sequencing forms the basis of our study.
