A new transgene competition system

Gene function screening is widely conducted using plasmid and viral vectors. Although both vectors are easy to use and convenient, a transient plasmid expression occurs unavoidable of uncontrollable copy number and stable cell lines by these two vectors have happened positional effect expression and disrupting the genes at the insertion site. For functional expression with reproducibility and competitive functional screening among introduced transgenes, it is desirable that expressions of all transgenes are stable and similar level. To address these issues, we applied our dual RMCE (recombinase-mediated cassette exchange) system, which we have developed an efficient gene insertion system with VCre/VLox and SCre/SLox recombination systems.
