We measured steady-state transcript levels in 1) wild-type C. crescentus, 2) an spdR response regulator deletion strain (ΔspdR), and an spdR phosphomimic strain (SpdR(D64E)) Overall design: Caulobacter crescentus wild type strain CB15, ΔspdR and spdR(D64E) strains were grown to mid log phase (OD600 ~0.3) in M2 defined medium containing 0.15% xylose. Cells were rapidly pelleted and resuspended in Trizol for RNA purification and subsequent RNA sequencing.
