Immunoglobulin A (IgA) protects mucosal surfaces against microbial infections. In response, many bacteria have evolved proteases that specifically target this antibody. Among them is the M64 IgA peptidase (IgAse), a ~140 kDa membrane-bound metallopeptidase produced by the gut commensal Thomasclavelia ramosa, which cleaves both IgA1 and IgA2 subclasses. Using small-angle X-ray scattering (SAXS), we characterized its flexibility and domain organization, and through cryo-electron microscopy (cryo-EM), we determined its structure at 2.8 Å resolution.
