This study aimed to develop macroporous bone scaffolds composed of a chitosan/agarose matrix reinforced with nanohydroxyapatite and enriched with stable CAPE to enhance their biomedical potential for applications in bone tissue engineering and regenerative medicine. A comparative biological evaluation of the fabricated nanohydroxyapatite-based bone scaffolds enriched with the two CAPE derivatives was performed using human osteoblasts through cytotoxicity, cell proliferation, and osteogenic differentiation assays. In addition, reactive oxygen species (ROS) and reactive nitrogen species (RNS) generation by human immune cells, as well as biofilm formation, were assessed. The scaffolds were also characterized in terms of microstructure, mechanical properties, and their surface apatite-forming ability. The folder ‘Microstructure characterization’ contains raw stereoscopic images, raw SEM images of the biomaterials, and raw results from microCT analysis, including images and porosity data. The file ‘Mechanical parameters’ contains results of compressive strength testing and measurements of Young’s modulus of the biomaterials. The file ‘Bioactivity assay’ contains raw SEM images of apatite deposits formed on the biomaterial surfaces, along with their EDS analyses. The folder ‘Cytotoxicity assessment’ contains raw CLSM images of cells on the scaffolds after live/dead staining, as well as raw MTT assay results. The folder ‘Cell proliferation assessment’ contains raw CLSM images of cells on scaffolds after AlexaFluor 635-conjugated phalloidin and DAPI staining, along with raw MTT assay results. The folder ‘Osteogenic differentiation’ contains raw results from ELISA assays and raw SEM images of cells on biomaterials after osteogenic differentiation experiments. The file ‘ROS/RNS Generation by Immune Cells’ contains raw results from experiments on ROS/RNS generation by neutrophils and monocytes. The folder ‘Evaluation of Biofilm Formation’ contains raw CLSM images showing fluorescent staining of biofilms formed by Staphylococcus aureus and Staphylococcus epidermidis on the biomaterial surfaces after 24 h of bacterial incubation.
