Dynamics of mutant hematopoietic progenitor cells in multiple myeloma and its association with clinical outcomes

TP53 mutant hematopoietic progenitor cells (HPC) from multiple myeloma (MM) patients may expand under lenalidomide-based regimens and give rise to secondary primary malignancies (SPM). Genetic screening may facilitate individualized treatment to minimize the risk of SPM. However, there are no longitudinal studies in purified HPC to inform on the evolution of gene mutations and its association with clinical outcomes. We sequenced CD34+ HPC sorted from MM patients all of whom treated with lenalidomide-based regimens.
