Activity-based protein profiling - Metagenomic datasets. hot springs metagenome

This study presents environmental activity-based protein profiling (eABPP) for function-driven enzyme discovery from natural communities. As a showcase, we identified active thermostable serine hydrolases using a combination of genome-resolved metagenomics and mass spectrometry-based activity-based Protein Profiling (ABPP). Accordingly, we performed metagenomic sequencing of DNA extracted from the epi-sedimentary communities of two hot springs, i.e. the 'Arkashin shurf' and the 'Helicopter spring', both located in the Uzon Caldera, Kamchatka Peninsula, Russia. Based on the raw metagenomic reads, we conctructed a metaproteome that served as a protein database for the identification of active proteins from the two communities that were labeled on site.
